Identifizierung neuer Tumorstammzell-basierter Therapieansätze beim humanen Gallenwegskarzinom

Therapeutic approaches for patients with biliary tract cancer (BTC) are limited and thus entail poor prognosis. New approaches combining chemotherapeutics are encouraging yet not able to overcome recurrence and lethal tumour progression. Cancer stem cells (CSCs) have been identified as the origin of cancer development and the cause of chemoresistance and recurrence. Based on these findings this project is designed to investigate whether CSCs represent promising targets for novel targeted therapies. In this project clinical specimens will be screened for expression of CSC markers and other factors including microRNAs which constitute the chemoresistant CSC phenotype. Furthermore, BTC cell lines will be used in order to develop a combination of chemotherapy and targeted therapies which inhibit embryogenesis-related signalling pathways or target epigenetic mechanisms. The results of this project enable development of new effective therapies for patients with biliary tract cancer.
Die limitierten Therapieoptionen für Gallenwegskarzinom-Patienten bedingen eine schlechte Prognose. Neue Chemotherapie-Kombinationen zeigen einen positiven Effekt, jedoch stellen Rezidive bzw. letale Tumorprogression nach anfänglichem Ansprechen ein großes Problem dar. In aktuellen Forschungen wurden Tumorstammzellen (CSCs) sowohl als Ursprung der Krebsentstehung als auch als Ursache für die Entstehung von Chemoresistenz und Rezidiven identifiziert. Wir prüfen in diesem Projekt, inwieweit CSC für neue zielgerichtete Therapien in Frage kommen. Dabei untersuchen wir in klinischen Proben die Expression von CSC-Markern sowie spezieller microRNAs, welche den Phänotyp chemoresistenter CSCs bestimmen. In Kombination mit Chemotherapie werden darauf basierend neue Therapien in vitro getestet, welche auf der Hemmung von Embryogenese-Signalwegen sowie epigenetischer Mechanismen beruhen. Die Daten dienen als Basis für die Entwicklung neuer effektiver Therapien für Patienten mit Gallenwegskarzinom.
